Why challenge your ELISA?

Challenging your assay for each new molecule is essential to selecting the most suitable kit.

We used two different kits to test nine commercial biologics for residual HCP.

Cytiva: Amersham HCPQuant CHO (Supernatant) Kit reported higher HCP in four samples.
Cygnus Technologies: CHO HCP ELISA 3G (F550) Kit reported higher HCP in two samples.
Both kits reported similar HCP in three samples.

We calculated the lower limit of detection (LLD) as the concentration where the signal is greater than three standard deviations from the mean of the zero standard [0.106 + (3 × 0.003) = 0.115].

We calculated the lower limit of quantification (LLQ) as the lowest concentration for which coefficient of variation (CV) is < 20% and recovery is between 80% and 120%. We determined the LLD and LLQ from eight replicates across two plates.

Reproducibility for robust assays and confidence in your results

We spiked the assay with three sample concentrations to perform precision analysis. We calculated intra-assay precision from 10 replicates for each concentration, and calculated inter-assay precision from the averaged means of three replicates (three plates).
